(Fiona and Shota are about to go to the park to practice for the Yokai Sumo Tournament. Suddenly, I heard the sound of heavy rain.)

I guess not.
基本的には 「そうではないと思うよ」と言うときのカジュアルな表現です。
ストーリーでは 「そうだね」 という意味で使われていました。
相手が否定的なことを言ったときに、 I guess not. と言うと、「そうだね」 という意味になりますよ。
ストーリーでは、 しょうたが We can’t train in the park today.
と言ったあと、フィオナが I guess not. と言っていましたね。
